หลังจากเหตุการณ์ Mansion Incident คริส และจิลล์ ก็ได้ออกไล่ล่า เปิดโปง Umbrella กันต่อไป เกี่ยวกับเหตุการณ์ที่เกิดขึ้นทั้งหมด

หน่วย S.T.A.R.S ได้ถูกสั่งพักงาน และถูกตราหน้าว่าเป็นอาชญากร เนื่องจากถูกกล่าวหาว่าสิ่งที่พวกเขาไปพบเจอมานั้น เป็นการสร้างเรื่องเท็จขึ้นมาโจมตี Umbrella เท่านั้น

Jill Valentine
Age : 24
Having undergone Delta Force training made Jill a natural fit as Rear Security for the Raccoon Police Department's special forces team, S.T.A.R.S., and her knack for picking locks and handling explosives have proven equally useful making her an especially outstanding team member. However, Jill is one of few surviving members after the team saw numerous losses to the shambling corpses and monstrous creatures they encountered while investigating reports of missing persons in what has become known as the Mansion Incident.
A survivor of that incident, Jill is determined to continue investigating Umbrella, the creators of the deadly T-Virus at the heart of the incident, but her investigation has been impeded by a suspension and an order to remain at home. What's more, her apartment appears to be under round-the-clock surveillance. The entire city seems to be under Umbrella's control, with the police force being no exception. Jill has decided to escape from her confines in Raccoon City in order to bring Umbrella's evil to light, but this may not prove so simple...

Carlos Oliveira
Age : 21
The assignment Umbrella gave was simple. U.B.C.S. was more than equipped for the task, being a collection of veteran mercenaries from around the globe. Carlos himself had fought countless battles as a guerrilla fighter—there's not a weapon he can't use, and few vehicles he can't pilot.
However, upon entering the city the platoon found a situation far more dangerous than the one on which they had been briefed. A mysterious virus was spreading at an unheard of rate, and the platoon was all but wiped out in less than two days.
Still, Carlos will not back down. His personal values and innate courage give him the strength to keep going, determined to rescue as many surviving citizens as possible. Time to head back out into the streets of Raccoon City and look for more survivors.

โดยการกลับมาครั้งนี้ เกมจะพาคุณดำดิ่งเข้าสู่การสืบสวน รวมถึงปมบางอย่าง สาเหตุของเรื่องราวทั้งหมดที่เกิดขึ้น ณ Raccoon City และการหลบหนีครั้งสุดท้ายของเธอ

-โดยภาคนี้ตัวเกมยังได้รับการพัฒนาจากขุมพลัง RE Engine (Reach for the moon) เฉกเช่นเดียวกับ Resident Evil 7, Resident Evil 2 และ Devil May Cry 5
-และตัวเกมได้กลับมาพร้อมกับมุมกล้องแบบ OTS (Over the shoulder)
-โดยช่วงต้นตัวเกมจะเป็นกึ่ง Open World เพื่อให้เราได้สำรวจเมือง Raccoon City พร้อมกับ Optional Area ที่จะทำก็ได้ หรือไม่ทำก็ได้ แต่เมื่อไปหอนาฬิกาแล้ว ตัวเกมจะเล่าเรื่องแบบเส้นตรงเพื่อให้ประสบการณ์ในแบบ Resident Evil 3 Original
